Comprehensive stratum corneum ceramide profiling reveals reduced acylceramides in ichthyosis patient with <i>CERS3</i> mutations

چکیده

The stratum corneum (SC) of the epidermis acts as a skin permeability barrier, and abnormalities in SC formation lead to several disorders. Lipids, especially epidermis-specific ceramide classes ?-O-acylceramides (acylceramides) protein-bound ceramides, are essential for barrier formation. Ceramide synthase 3 (CERS3) is involved synthesis acylceramides CERS3 mutations cause autosomal recessive congenital ichthyosis. In present study, we measured activity performed comprehensive profiling an ichthyosis patient with compound heterozygous mutations: nonsense mutation p.Arg75* missense p.Arg229His. p.Arg229His mutant proteins was reduced 4% 56%, respectively, wild-type protein. patient’s SC, acylceramide levels were greatly reduced, but ceramides remained almost unchanged. Non-acylated also affected patient; particular, composed sphingosine non-hydroxy or ?-hydroxy fatty acid substantially higher than healthy controls. These results suggest that reduction alone leads Although synthesized from acylceramides, not necessarily correlated.

Characterization of overall ceramide species in human stratum corneum.

Ceramides (CERs) in human stratum corneum (SC) play physicochemical roles in determining barrier and water-holding functions of the skin, and specific species might be closely related to the regulation of keratinization, together with other CER-related lipids. Structures of those diverse CER species, however, have not been comprehensively revealed. The aim of this study was to characterize over...

LC/MS analysis of stratum corneum lipids: ceramide profiling and discovery.

Ceramides (CERs) in the upper layer of the skin, the stratum corneum (SC), play a key role in the skin barrier function. In human SC, the literature currently reports 11 CER subclasses that have been identified. In this paper, a novel quick and robust LC/MS method is presented that allows the separation and analysis of all known human SC CER subclasses using only limited sample preparation. Bes...

Desquamation in the stratum corneum.

To maintain a constant thickness of the stratum corneum the desquamation rate and the de novo production of corneocytes is delicately balanced. Using a plantar stratum corneum model we have obtained evidence that proteolysis is a central event in the desquamation process. A number of regulatory mechanisms for desquamation have been postulated based on our findings.

Stratum corneum dysfunction in dandruff

Dandruff is characterized by a flaky, pruritic scalp and affects up to half the world's population post-puberty. The aetiology of dandruff is multifactorial, influenced by Malassezia, sebum production and individual susceptibility. The commensal yeast Malassezia is a strong contributory factor to dandruff formation, but the presence of Malassezia on healthy scalps indicates that Malassezia alon...

Skin diseases associated with the depletion of stratum corneum lipids and stratum corneum lipid substitution therapy.

The skin is the largest organ of the body, whose main function is to protect the body against the loss of physiologically important components as well as harmful environmental insults. From the inside to the outside, the skin comprises three major structural layers: the hypodermis, the dermis and the epidermis. The epidermis contains four different sublayers, the stratum corneum (SC), stratum g...
